Purpose:

To determine the safe dose of intravitreal azithromycin in rabbit eyes
|
Methods:
|
Fourteen albino rabbits were scheduled for evaluating side effects of seven intravitreal doses of 100, 200, 300, 400, 500, 1000, and 2000 micrograms (µg)/0.1 cc of azithromycin, two rabbits for each dose. For each dose (two rabbits), the drug was injected into the right eye and balanced salt solution was injected into the other eye. All eyes were examined in 1, 2, 3, 7, 14 and 28 days after intravitreal injections. Electroretinogarphy (ERG) was performed before and 28 days after intravitreal injection. After four weeks, the animals were euthanized and eyes were enucleated and submitted for haematoxylin and eosin and immunohistochemistry evaluations.

Results:
|
Persistent posterior subcapsular opacity developed in 2 eyes in 2000 µg and one eye of 1000 µg dose. In histopathology, the integrity and the appearance of all retinal layers were preserved in 100, 200, 300, and 400 µg doses and in control eyes. Severe atrophic changes of all retinal layers together with cystoid degeneration and formation of a disorganized thin retina were observed in 2000, 1000 and one of 500 µg/0.1ml injected eyes.

Conclusion:
|
Intravitreal injection of azithromycin in rabbit eyes was not associated with adverse toxic effects up to 400 µg doses.
