Purpose:
|
The ADAMTS family has an important role in extracellular matrix (ECM) degradation and turn over. One of the potential contributors to AMD pathology is retinal pigment epithelial (RPE) cell senescence. With extensive passaging, continuous accumulation of products of oxidation such as ROS, protein carbonyls and lipofuscin has been found in senescent RPE grown in vitro. The accumulation of oxidative damage is one of the most widely accepted causes of ageing. Here we report the effects of senescence on the expression of ADAMTS 5, 15 genes.

Methods:

Cultured primary RPE cells entered replicative senescence after serial passages. First strand cDNA synthesis from 1000 ng of total RNA was performed by random hexamers. Real time PCR was performed using the Quantity Fast SYBR Green PCR Kit. Fold change in expression were obtained using software provided by the Corbet instrument.

Results:
|
Our results show that the ADAMTS 5, 15 genes are expressed in the RPE cells and their expression in senescence cells are reduced in compared with normal cells.

Conclusion:
|
Accumulation of ROS in RPE cell senescent has effect on expression of ADAMTS 5, 15 genes. Our finding indicates that these genes may have potential roles in age-related macular degeneration pathogenesis.
