Purpose:
|
The ADAMTS family has an important role in extracellular matrix (ECM) degradation and turn over. Oxidative damage in the RPE cells is due to an imbalance between the generation and the elimination of reactive oxygen species (ROS) which causes RPE cells dysfunction. This is a major cause of ECM damage in AMD. The purpose of this study was to investigate the effect of oxidative stress on the expression of ADAMTS 1, 3 genes in RPE cells.

Methods:

Cultured human RPE cells were subjected to oxidative stress induced by 100, 250, 350 µM H2O2. Cell viability was measured using the microculture tetrazoline test (MTT) assay. Changes in gene expression were measured by a Real time PCR assay.

Results:
|
H2O2 decreased cell viability compared to the control level. Oxidative stress affected on expression of genes and expression of ADAMTS 1, 3 genes reduced.

Conclusion:
|
Oxidative stress has some effects on the expression of ADAMTS 1, 3 genes in RPE cells. Our finding indicates that these genes may have potential roles in age-related macular degeneration pathogenesis.
